Definition
CYLD ist eine Ubiquitin-Carboxyl-terminale Hydrolase. Sie entfernt Lysin- und lineare Ubiquitin-Ketten von Zielproteinen. CYLD reguliert u.a. NF-κB-, JNK-, Wnt-, TGF-β- sowie Notch-Signalwege negativ und beeinflusst dadurch die Zellproliferation, Apoptose und Entzündungsprozesse.
Genetik
CYLD wird durch das gleichnamige Gen kodiert, das sich auf Chromosom 16 am Genlokus 16q12.1 befindet.
Funktion
Neben der oben erwähnten Hauptfunktion als deubiquitinierendes Enzym, ist CYLD an der Aufrechterhaltung der Zellhomöostase, der Ziliogenese und der Regulation von T-Zellen beteiligt.
Darüber hinaus ist es in der postsynaptischen Dichte von Neuronen angereichert und spielt eine wichtige Rolle bei der neuronalen Morphologie und Plastizität. Es beeinflusst das Wachstum von Dendriten und die Reifung von postsynaptischen Spines, unter anderem durch Modulation der Tubulin-Acetylierung und Interaktion mit AMPA-Rezeptoren.
Klinik
Mutationen im CYLD-Gen führen u.a. zum autosomal-dominant vererbten Brooke-Spiegler-Syndrom. Die Erkrankung ist durch die Entwicklung multipler, meist benigner Tumoren wie Zylindromen, Spiradenomen und Trichoepitheliomen gekennzeichnet, die bevorzugt an Kopf und Hals auftreten. Die Pathogenese umfasst einen „Zwei-Treffer“-Mechanismus, bei dem die biallelische Inaktivierung von CYLD in Tumorzellen zu einer Dysregulation der NF-κB-Signalübertragung und einer erhöhten Zellüberlebensrate führt.
